Optimization of process parameters for alkaline protease production under solid state fermentation by alkalophilic Bacillus Sp

Abstract

Production of extracellular alkaline protease employing our laboratory isolated bacterial species, Bacillus Sp. (PE-11) under solid state fermentation was optimized. Different substrates like wheat bran, green gram bran, black gram bran, corn flour, barley, maize bran, rice bran, jowar flour, wheat rawa and sugar cane bagasse were studied to optimize the best substrate. Wheat bran showed the highest activity. The physical and chemical parameters were optimized. The maximum enzyme activity under optimum conditions was 178,0 PU/g of wheat bran. The optimum conditions are incubation time of 72 h, incubation temperature at 37°C, glucose as additive 1% w/w, corn steep liquor as additive 1% w/w, inoculum level 10% v/w, moisture content of solid substrate 70%, 1:50 ratio of substrate weight to flask volume and pH 10.0.
