Identification of Oryza sativa × Oryza officinalis F1 and Backcross Progenies Using Genomic in situ Hybridization

Abstract

With digoxigenin-labelled total genomic DNA from O. officinalis as probe and 16-fold unlabelled total genomic DNA from O, sativa as blocking, genomic in situ hybridization was applied to somatic chromosome preparations of Oryza sativa × Oryza officinalis F1 hybrid and backcross progenies, which led to the confirmation of three double and four monosomic alien addition lines, When anti-digoxigenin-POD and DAB were used for detection, the color of officinalis chromosome was brown, against light blue sativa chromosome from Giemsa counterstain. When anti-digoxigenin - FITC was used instead, officinalis chromosome appeared greenish- yellow while sativa chromosome fluoresced red with PI counterstain. In the interphase of alien addition line, officinalis chromatins were visualized as distinct areas. Hence it is likely to be also feasibie for discriminating genome A from other oryza genomes.
