Analysis of microarray titration experiments

Abstract

In this paper we introduce a novel approach for the analysis of microarray titration experiments. The test introduced here can be used more generally for drug-dosage effect or time effect for a drug treatment. It is based on a multivariate version of Fieller's theorem for testing ratios of normal means. The procedure is illustrated using data on mixtures of RNA samples from the MAQC project. Different background-correction methods and normalization methods are compared using this approach. Titration samples, obtained by mixing known proportions of independent samples, are important in the study of reliability and consistency of the microarray technology.
