A two-stage sib-pair genome scan for schizophrenia susceptibility genes

Abstract

We have undertaken a systematic search for linkage in 200 affected sibling-pairs (ASPs) with DSM-IV schizophrenia using semiautomated genotyping with fluores-cent labelled microsatellite markers. In Stage 1 we typed 220 markers at approximately 20 cM intervals in 100 ASPs. Single locus sib-pair analysis was carried out using SPLINK and multi-locus analysis using MAPMAKER SIBS. This identified seven regions of interest with an MLS > 0.74 which corresponded to P < 0.05 from simulation studies. These regions were selected for genotyping in Stage 2 in which a total sample of 197 sib-pairs, including parents and unaffected siblings where available, were genotyped with 76 microsatellite markers with an average marker density of 5.1 cM. Analyses of these data are currently being completed and the order of Stage 2 markers being checked by radiation hybrid mapping.
